Spojuje námořní přístav Brest ve Francii u Atlantiku s ruskou Machačkalou na pobřeží Kaspického moře. V blízkosti se nachází 50. rovnoběžka severní šířky. V Česku tvoří hlavní západovýchodní silniční osu (dálnice D5, většina dálnice D1).
